Zymograms to analyze catalytic activity of fusion enzymes. SDS-PAGE of purified single and fusion enzymes were loaded after denaturation (incubation at 70°C for 5 min in SDS-containing buffer). Protein bands were detected with silver staining. In case of in-gel activity assays, enzymes were washed with Triton X-100 and incubated at optimal conditions in reaction buffer. Utilization of soluble CMC-cellulose was used to detect the activity of endoglucanase, while esculin and X-Gal were applied to visualize the catalytic performance of β-glucosidase.
